Angiostop attacks cancer at both the cellular membrane as well as the DNA level. At the cellular membrane level, it inhibits new blood vessel formation and four different RTKs: vascular endothelial growth factor receptor (VEGFR), platelet-derived growth factor receptor (PDGFR), epidermal growth factor receptor (EGFR), and fibroblast growth factor receptor (FGFR). Simultaneous suppression of these RTKs as well as its associated downstream signaling (e.g. Akt, FAK, Paxillin, Erk) appears to be more effective than suppression of a single RTK. is is because each RTK is associated with one or more cancer process. For example, angiogenesis (or new blood vessel formation) is associated with VEGFR, PDGFR, and FGFR. Apoptosis (or cancer cell death) is associated with EGFR and VEGFR. Metastasis is mainly associated with FGFR and so on. It is clear that the more RTK targets that are addressed, the higher the chance of completely suppressing associated cancer processes. For example, drugs like Sutent and Nexavar, which inhibit both VEGFR and PDGFR, can address several cancer processes while a drug like Gleevec can only target proliferation and tumor vascular maturation. Or a drug like Tarceva can target apoptosis, mitogenesis and cellular motility only, not all of the processes. None of the drugs target FGFR. Angiostop, on the other hand, inhibits all four RTKs, so it is able to block more cancer processes. Revivin Revivin is considered a G1-S inhibitor, recommended as an adjunct cancer therapy.
